| Argonaute(Ago)蛋白是不受原间隔相邻基序(Protospacer Adjacent Motif,PAM)限制的可编程核酸酶,其催化过程简便且易于精确调控。该蛋白具有成本低、操作简单等多方面的优势,在多重检测、原位分析及模块化设计等方面具有显著的技术潜力。本文首先简单阐述了Ago蛋白结构及催化分子机制,综述了基于Ago蛋白的生物传感技术的研究进展,总结了其在细菌、病毒、真菌及体液中的核酸检测方面的应用,并系统性分析了该技术在应用过程中所面临的问题和挑战。 |

| Argonaute (Ago) proteins are programmable nucleases that are not restricted by the Protospacer Adjacent Motif (PAM), featuring straightforward catalytic processes and precise controllability. These proteins offer advantages such as low cost and operational simplicity, demonstrating significant technical potential in multiplex detection, in situ analysis, and modular design. This review first outlines the structural domains and catalytic mechanism of Ago proteins. It then surveys recent progress in Ago-based biosensing strategies, summarizing their applications in detecting bacterial, viral, and fungal targets, as well as in analyzing nucleic acids within body fluids. Finally, the article provides a critical discussion of the current challenges and future perspectives for the field. |
